Knowing that the cob is likely to crack as it dries, is it better to build the structure all in one go or in layers, letting the latest one dry before adding the next?

Well.. Cob doesn't HAVE to crack when it dries.. As a general rule, if it does you haven't added enough sand, though more straw can fix it too.
If you can, you want to build it up in one monolithic chunk. If you have to do layers, it's best not to let each layer dry completely but only enough that it will hold the next layer without sagging. Ideally, you want to be able to knead the layers together into one solid mass.
If the layers dry completely, it can be difficult to stick the next layer on which can cause separation and MORE cracking.
